I need to upload a large number of user so they can be used inSharePoint. What is the best way to do this so that I can take aspreadsheet and upload? Also, I would like to assign groups.

You can grant any Domain groups like NT AUTHORITY\Authenticated Users, alsoyou can retrive any mailing list from your outlook or whatever.
If you have a list of names (email addresses) in a text file, one perline, you can just paste the list in. Same thing with groups.
If your users are associated with some common domain then you can make somegroups on that domain and then those users can be added to particular group andthere you are just give permission to the group and it would be scalable too.
