SharePoint and .Net

Does anybody has experience with SharePoint in combination with .NET? We
like to build our own webparts with ASP.NET. Just "adding" aspx-files to
the portal/resources-folder gives a permission denied (asp.net filters
are enabled on the webserver).

i am also facing same problem,
how can i create web part in asp.net is share point
server support .net framwork becouse i think for
execution of aspx file you should need .net framwork ,
is this inbuild in share point ..Please let me know as
soon as possible

i have also tried these option but still it's giving
me same error
error is here
Access denied  to
'\\.\backofficestorage\localhost\SharePoint Portal
Server\workspaces\sample\Portal\' directory. Failed to
start monitoring directory changes.
