I would recommend creating a new wep app for the new address and
extending an existing one. That has the best chances of making the
changes you need in IIS and SharePoint. Besides that you should be
fine.

DO NOT delete the original Web app after
you have extended it to the new port  number. Doing so will break the entire set
of Web Applications.

If you've already created the application, changing the port  number is
touchy. You'd do better to create a new SP application through Central
Admin or extend your current application.

You can change  the port  in IIS, modify the host header in
IIS (if necessary) and also modify the default AAM in Central Admin. But, the
IIS change will not be recorded in the configuration database. Therefore, if you
add/re-add the Windows sharepoint  Services 3.0 server role, you will get the old
settings and must 're-do' the IIS changes on that server.

(make a backup before this...) You can also delete the Web application,
re-create using your existing databases and get the same effect. This will now
be written in the configuration database (this is what peter is referring to)
